They are keeping awfully tight lipped on this. Would be nice to know a little bit more about it, I think the last details we were given was what we got from E3.
No kidding, there's too much talking in Mass Effect for it to work with W40K. W40K is all about brutality and survival, not long philosophical rants.
Still if you put the Inquisition in a 40k game it would probably make an excellent RPG. They did go a bit overboard in Mass Effect with all the talking, I know, but include a better fighting system and it would make for a very good game. Space Marines just aren't the right types for a game that is mostely role playing.

Space Marines are all about killing and asking questions later, the Inquisition is more the other way around. Even the Eldar or Tau would work better for an RPG.

An eldar rpg would be interesting, Start the game as a guardian, and depending on the way you complete your missions and the way you act in the game world, you start getting bonuses and upgrades that start leaning towards one of the aspect temples, going the path of the seer, going the path of the loner (ranger), or for those who stay closer to the middle, get boosted in rank withing the normal guardians, (guardians, jebike (shining spear path available after guardian jet bikes), Storm Guardians,(Dire avengers open here), autriach, and at any point before autriarch is when you can start going to a specific path instead of becoming an autriarch) the game would have many possibilities!

That's the only thing I can see in the W40K universe as working in the RPG role. Orks wouldn't work, IG wouldn't really work, and SM definitely wouldn't work.

IG could have your standard trooper advancing up the ranks say to a Sergeant and beyond or a depending on what you liked doing, a Commisar or a heavy weapons team member.
SM I guess you could have a scout working his way up to become a full space marine up to captain status.
Orks you could be a gretchin and eventually work your way up to become a nob or boss.
Chaos you start off as a Cultist and work your way up to a chaos marine and depending on what chaos power you choose you could become a chaos lord or a sorcerer.
I can only see those as working in the sense of leveling up and getting upgrades. However, I don't think they would be that great for dialogue and storylines. Because a guardsman doesn't get options, he just does as he's told. A cultist is pretty much the same way and a marine does the same as a neophyte. And Orks just fight and fight, there's no dialogue regardless.
