I was going to answer with KotOR. Because that was a huge game, and one of those rare games where I thought, 'I need more of this', so the instant I'd finished it I started a new game. And then another new game. And another. We're talking hundreds of hours here, if not thousands.
And then I realised that the Mass Effect series, in total, has probably claimed more hours as a whole. Thousands is a minimum here, rather than an upper estimate.
Then I realised that if we're including entire franchises, that The Elder Scrolls probably holds the record.
And then I realised that the Fallout series probably has more hours under my belt than even that.
And then I realised, fuck you. I'm a nerd. I can't quantify. And Vampire; Bloodlines: The Masquerade probably holds the record.
Diablo 2. I put countless hours of my life into that game over the course of a six or seven year period, thousands of Baal runs and play-throughs on Hell difficulty, but never once getting IK armour in a drop... That game used to be great, but over time there developed just a handful of character builds that were actually useful in Hell, like Hammerdins, and IMO the game became fairly boring as there was no longer much variety in character builds and gameplay.

Probably Mass Effect. I would guess the total number of hours combined from all three games in the series would have to be around 250. And that's not even including ME3 multiplayer which i've spend endless hours on...

I spent roughly 2000 hours playing Star Wars Galaxies over the years. Jedi Academy is probably in the hundreds at least. I have spent probably about 500 hours on the Mass Effect Trilogy too, i usually take about 70 hours per game.
Back when I played WoW I spent thousands of hours leveling up and grinding for things.
Now that I play STO I've probably also spent thousands of hours leveling up and I've spent even more time recently grinding the elite stf's to get the MK XII M.A.C.O. gear. I'm now working on grinding those same elite stf's to grind the MK XII Omega Force gear. I'm also running missions that grant fleet marks to give to the growth of our fleet starbase.
